The sterilization principle of nisin is primarily based on its destructive effects on bacterial cell membranes. It is a natural antimicrobial peptide produced by Lactococcus lactis, with a broad spectrum of antibacterial activity. Nisin mainly disrupts the integrity of bacterial cell membranes by interfering with the biosynthesis of peptidoglycan in the bacterial cell wall and affecting the synthesis of membrane phospholipids. When nisin comes into contact with bacterial cell membranes, it can form tiny pores or channels within the membrane, leading to the rapid leakage of essential intracellular substances such as ATP, amino acids, and other ions while allowing extracellular water molecules and other small molecules to enter the cell. This alteration in membrane permeability causes autolysis of the cell, nutrient loss, and a decrease in membrane potential, ultimately resulting in the death of pathogenic and spoilage bacteria.

Nisin exhibits significant antibacterial effects against Gram-positive bacteria, especially some resistant strains. It can effectively inhibit or kill these bacteria, thereby extending the shelf life of food products. However, its antibacterial effectiveness is weaker or absent against Gram-negative bacteria, yeasts, and molds. This may be related to the complexity of the cell membranes of Gram-negative bacteria and the protective role of their outer structures, such as the lipopolysaccharide layer.

Due to the natural, safe, and non-residual properties of nisin, it is widely used in food preservation, including dairy products and meat products, to prolong shelf life and maintain quality. Additionally, it has potential medical applications, such as in the preparation of antibacterial drugs and disinfectants. However, its application is also limited by factors such as weaker antibacterial effects against Gram-negative bacteria and stability issues under certain conditions.

This mechanism makes nisin an efficient, non-toxic, and safe natural food preservative, with broad application prospects in the food industry.
