Nisin factories primarily employ the following processes for the production of nisin:

1.Strain Cultivation: First, high-producing strains of lactic acid bacteria are selected for pre-cultivation. The strain is introduced into a culture medium containing suitable nutrients, and cultivation conditions such as temperature, pH value, and oxygen supply are adjusted. The strain is cultured for a specific time (usually 12 to 24 hours) to grow to an appropriate stage.

2.Fermentation Process: The cultivated strain is inoculated into fermentation vessels and an appropriate fermentation substrate, such as sucrose or lactose, is added. Fermentation conditions are adjusted, including temperature, pH value, stirring speed, and oxygen supply, to promote the growth of lactic acid bacteria and the production of nisin. The fermentation process generally lasts from several days to several weeks.

3.Separation and Purification: After fermentation, the fermentation broth is processed for separation and purification. Initially, methods like centrifugation separate the bacteria and fermentation waste liquid, obtaining a liquid containing nisin. Subsequently, the nisin is further purified through filtration, concentration, and washing processes to remove impurities and harmful substances, yielding high-purity nisin.

4.Drying Formulation: The purified nisin is then dried, commonly using methods such as spray drying or freeze-drying. The dried nisin can be used to prepare various products.

Additionally, some factories utilize specific processes to enhance the extraction yield of nisin, such as adjusting the pH and temperature of the fermentation broth to adsorb nisin onto a medium, followed by steps like solid-liquid separation, acidic water treatment, and further solid-liquid separation, ultimately producing the nisin product. These processes can effectively improve the extraction efficiency of nisin and reduce waste liquid production.

The specific production process may vary depending on the factory and equipment. During production, it is crucial to strictly control the parameters and conditions of each stage to ensure the quality and safety of the product.
